Molecular Analysis of Medically and Veterinary Important Muscid Flies (Diptera: Muscidae) in Thailand

چکیده
We demonstrated the using of the internal transcribed spacer (ITS2) of ribosomal DNA as a tool for identification of medically and veterinary important Muscidae flies in Thailand. A total of 27 fly samples were collected from various regions of Thailand. Six fly species in three subfamilies including Azeliinae (Hydrotaea spinigera), Muscinae (Musca domestica, M. sorbens) and Stomoxyinae (Stomoxys calcitrans, S. indicus and S. sitiens) were identified base on morphological taxonomy. PCR amplicons of the ITS2 gene of these flies varied between 312-377 bp with A+T content of 76.6%. ITS2 sequences of the flies in this study were 93-100% identity to sequences in database and 21 samples were compatible with morphological identification, while sequences of 6 samples did not match any sequences in the database. The intraand inter-specific divergence analysis results showed that the maximum of intra-specific (within species) variation (6.9%) was found in M. domestica while the minimum inter-specific (between species) variation (11.9%) was found in the sister grouped couple of S. sitiens and S. indicus. No overlapping between intraand inter-specific divergences was found in all species of this study. The bootstrapped NJ tree constructed showed ability to classify each subfamily in to monophyletic clades. PCR-RFLP using XapI restriction enzyme digestion was able to differentiate between the three Stomoxys species. Data obtained from this study would be valuable for both medical and veterinary entomologists for more accurate identification of important fly species. Therefore, it could be used for population dynamics studies and enrolled in integrated pest management control program.

Background and Aim: Some medically important species of Muscidae and Fanniidae families are among synanthropic flies and can be seen in human habitats therefore they can be mechanical vectors of different pathogenic microorganisms. Identification and determination of species of these flies is essential for any control program planning. This study was conducted to determine the faunal diversity ...
متن کاملInsect arrival pattern and succession on buried carrion in Michigan.
This study examined pig carcasses buried at two different depths, 30 and 60 cm, to determine if insects were able to colonize buried carcasses, when they arrive at each depth, and what fauna were present over seven sampling dates to establish an insect succession database on buried carrion in East Lansing, MI. Thirty-eight pigs were buried, 18 at 30 cm and 20 at 60 cm. Four control carcasses we...
متن کاملRate of development of Hydrotaea rostrata under summer and winter (cyclic and constant) temperature regimes.
The black carrion fly Hydrotaea rostrata Robineau-Desvoidy (Diptera: Muscidae) is a muscid, which occurs on approximately one-third of decomposing human corpses involved in homicide cases in south-western Australia. Work to date on its development rates is scant with only one published source available. The current study measured the precise developmental rates of this species using high repeti...
متن کاملStudy of Fauna and Biodiversity of Medically Important Flies in Juybar, North of Iran
Background and purpose: Flies have a high diversity and are able to transmit many pathogens due to their adaptation and close relationship with humans. Therefore, this study was performed to identify the fauna of medically important flies in the city of Joybar in northern Iran. Materials and methods: This descriptive-analytical study was conducted twice a month in Juybar at selected sites, inc...
متن کاملA Modified Trap for Adult Sampling of Medically Important Flies (Insecta: Diptera)
BACKGROUND Bait-trapping appears to be a generally useful method of studying fly populations. The aim of this study was to construct a new adult flytrap by some modifications in former versions and to evaluate its applicability in a subtropical zone in southern Iran. METHODS The traps were constructed with modification by adding some equipment to a polyethylene container (18× 20× 33 cm) with ...
